A shortcut method for sexing juvenile European sea bass, Dicentrarchus labrax L. ArchiMer

The objective of this study was to develop a rapid and effective method of sexing juvenile sea bass with minimum labour and material. To this end, the gonad squash mount technique was applied along with macroscopic techniques for sexing a large number of experimental fish at the age of 215-275 days post fertilization (p.f.). At this age, 90% of the 3894 fish could be unambiguously sexed by macroscopic examination of their gonads, whereas the remaining proportion was identified using squash mount preparations (10%). The accuracy of the observations was measured up to classical histology procedures. Undifferentiated fish accounted for 0.1% of the total population only. Affecting cucurbit hormonal status by a ZYMV transient expression vector National Institute of Agronomic Research

Squash and cucumber plants were infected with ZYMV strains engineered to over-express enzymes involved in ethylene and gibberellin biosynthesis. Presence of the inserted ACS gene within the viral RNA was transient and the insert was deleted after 20-40 days. Ethylene over-expressing plants produced very high ethylene levels and were severely stunted. Monoecious cucumbers produced earlier, consecutive female flowers. Gibberellin over-expression was deduced from the observed phenotypic change, plants were taller and flowered earlier.

Abstract: The objective of this work was to evaluate the evolutionary dynamics of the wild-weedy-domestic gene pool of Cucurbita argyrosperma squash by estimating the levels of genetic diversity and gene flow in the putative area of its domestication. Nine populations were collected, and DNA was extracted from young leaves harvested separately from approximately 20 individuals in each population. The DNA fragments were amplified with eight pairs of SSR primers and separated by electrophoresis in 5% denaturing polyacrylamide gels. Genetic diversity and the amount of gene flow were estimated in the populations, and Bayesian grouping was used to determine the levels of gene infiltration and probability of ancestry. Turkey is very rich in cucurbit genetic resources due to its diverse geographical and ecological situation. Turkey is one of the important diversity centers for cultivated cucurbits because of the adaptation to diverse ecological conditions as result of natural selection and also the farmers selection in accordance with their preference. National collection of Cucurbitaceae species at National Gene Bank of Turkey (AARI) consists of 2223 accessions. The tropical pumpkins Cucurbita moschata Duchesne and C. argyrosperma Huber are often found growing together. Both C. argyrosperma subsp. argyrosperma (the domesticated cushaw squash) and the wild species C. argyrosperma subsp. sororia (L.H. Bailey) Merrick & Bates are found together in Central America. Several reports indicate that plants displaying a phenotype combining traits of both species occur, suggesting gene flow between species. Response of hybrid squash Tetsukabuto to water depths and doses of nitrogen Horticultura Brasileira

ABSTRACT The aim of this work was to evaluate, under the Cerrado conditions of Central Brazil, the effect of different amounts of water and nitrogen (N) on the production performance of hybrid squash Tetsukabuto. Five water depths were tested in 2008 (185, 265, 343, 408 and 477 mm) and four in 2009 (286, 360, 430 and 540 mm), using line source sprinkler irrigation. The doses of N (0, 50, 100, 150 and 200 kg/ha) were the same in both experiments. The experimental design was a split-plot strip block with three replications. Fruit productivity was affected by the interaction between water depths and doses of N. Longitudinal striping is a common fruit color pattern in squash, pumpkins, and gourds of Cucurbita pepo, but cultigens can exhibit different kinds of fruit striping. Often, the stripes are broad and contiguous, but in some cases they are narrow and non-contiguous (“broken”) or, much more rarely, they can have an irregular distribution. These three striping phenotypes have been described and compared, and have been found to be under the control of alleles at the l-1 locus. Ordinarily, dark stripes appear between the 10 main subepidermal vascular tracts with the background color covering the rest of the fruit. Methyl bromide is a critical soil fumigant used in the production of several fresh fruit and vegetables grown in the U.S. The U.S. Clean Air Act of 1992, as amended in 1998, requires that methyl bromide be phased out of use by 2005. A mathematical programming model of the North American vegetable market indicates that the elimination of methyl bromide will have significant impacts on U.S. growers of fruit and vegetables that rely on methyl bromide for soil fumigation purposes. The schedule for eliminating methyl bromide has resulted in a 50% decline in methyl bromide availability and has resulted in significant increases in the price of methyl bromide.
