Resumo: O trigo (Triticum aestivum L) constitui uma das principais culturas de importância agrícola para o Brasil. Contudo, a ocorrência de doenças como a brusone, tem limitado a produtividade dessa cultura nas regiões produtoras. Desse modo, o objetivo desse trabalho foi avaliar o potencial de uma linhagem Streptomyces sp. In an attempt to screen out new potent antibiotic producers from soil, Streptomyces hygroscopicus D1.5 was isolated and was found antagonistic to both bacteria and fungi. Streptomyces is a genus of Gram-positive bacteria that grows in various environments, and its shape resembles filamentous fungi. The morphological differentiation of Streptomyces involves the formation of a layer of hyphae that can differentiate into a chain of spores. The most interesting property of Streptomyces is the ability to produce bioactive secondary metabolites, such as antifungals, antivirals, antitumorals, anti-hypertensives, immunosuppressants, and especially antibiotics. Antimicrobial activity of actinomycetes and characterization of actinomycin-producing strain KRG-1 isolated from Karoo, South Africa BJPS

In the present study we reported the antimicrobial activity of actinomycetes isolated from aridic soil sample collected in Karoo, South Africa. Eighty-six actinomycete strains were isolated and purified, out of them thirty-four morphologically different strains were tested for antimicrobial activity. Among 35 isolates, 10 (28.57%) showed both antibacterial and antifungal activity. The ethyl acetate extract of strain KRG-1 showed the strongest antimicrobial activity and therefore was selected for further investigation. This work aimed a survey on the biodiversity of maize endophytic actinomycete, and an evaluation of their potential to control the phytopathogenic fungi. From several regions of São Paulo state, 40 strains were isolated from the healthy maize plants. The identification of these strains, based on morphological properties and fatty acid methyl ester (FAME) profile showed that most of them belonged to the Streptomyces genus. Carbon catabolite repression of retamycin production by Streptomyces olindensis ICB20 BJM

Retamycin is an anthracyclinic antitumoral complex produced by Streptomyces olindensis ICB20. In this work the influence of different glucose concentrations in the feed medium on the production of retamycin was studied. Chemostat cultures employing glucose concentration varying between 10 g/L and 25 g/L showed that use of high glucose concentration resulted in catabolite repression of the biosynthesis of the antitumoral. The highest specific retamycin production rate, qRTM = 7.8 mg/g.h, was obtained when glucose concentration was 10 g/L. The lowest value of qRTM, 2.5 mg/g.h, was observed when glucose concentration was 20 and 25 g/L. Draft genome sequence of Streptomyces sp. strain F1, a potential source for glycoside hydrolases isolated from Brazilian soil BJM

ABSTRACT Here, we show the draft genome sequence of Streptomyces sp. F1, a strain isolated from soil with great potential for secretion of hydrolytic enzymes used to deconstruct cellulosic biomass. The draft genome assembly of Streptomyces sp. strain F1 has 69 contigs with a total genome size of 8,142,296 bp and G + C 72.65%. Preliminary genome analysis identified 175 proteins as Carbohydrate-Active Enzymes, being 85 glycoside hydrolases organized in 33 distinct families. This draft genome information provides new insights on the key genes encoding hydrolytic enzymes involved in biomass deconstruction employed by soil bacteria.

Effects of Brassica napus seed meal amendment on soil populations of resident bacteria and Naegleria americana, and the unsuitability of arachidonic acid as a protozoan-specific marker OAK

Bacterial and protozoan populations were monitored in orchard soil amended with Brassica napus seed meal. Numbers of fluorescent pseudomonads peaked one week following incorporation of the amendment and declined thereafter, whereas populations of Streptomyces spp. increased to stable levels nearly 20-fold higher than in non-treated soil. The amoeba-flagellate Naegleria americana, isolated from the amended soil, showed a marked feeding preference in culture for fluorescent pseudomonads relative to Streptomyces spp. Amended soils showed rapid declines in RSM-derived cis-oleic acid that paralleled increases in the numbers of bacteria and proportion of bacterial fatty acid markers in the soil. Este trabalho teve como objetivo avaliar o efeito de seis isolados de estreptomicetos na mortalidade e eclosão de juvenis de segundo estádio (J2) de Meloidogyne incognita e no controle da meloidoginose em mudas de tomateiro. Foi montado um bioensaio em placas tipo Elisa, sendo adicionados em cada célula, 200 µL de metabólitos dos isolados, com 20 µL de uma suspensão com 25 juvenis de segundo estádio (J2) de M. incognita. Os metabólitos produzidos por Streptomyces griseus subsp. griseus causaram 98,2% de mortalidade dos J2 de M. incognita. Em outro bioensaio, foram adicionados 3 mL dos metabólitos em frascos de vidro, com 100 µL da suspensão contendo 25 ovos de M.incognita. Evaluation of antimicrobial activity of the endophytic actinomycete R18(6) against multiresistant Gram-negative bacteria Anais da ABC (AABC)

Endophytic actinomycetes are promising sources of antimicrobial substances. This study evaluates the activity of metabolites produced by the endophytic actinomycete R18(6) against Gram-negative bacteria multiresistant to antimicrobials. R18(6) isolate was grown in submerged cultures under different conditions: carbon source, temperature, pH and incubation time to optimize antimicrobials production. The actinomycete grown in base medium supplemented with 1% glucose, pH 6.5 and incubation at 30 ºC for 96 h with shaking at 100 rpm, exhibited the highest activity against the used Gram-negative bacteria. Exploring plant growth-promotion actinomycetes from vermicompost and rhizosphere soil for yield enhancement in chickpea BJM

Abstract The main objective of the present study was to isolate and characterize actinomycetes for their plant growth-promotion in chickpea. A total of 89 actinomycetes were screened for their antagonism against fungal pathogens of chickpea by dual culture and metabolite production assays. Four most promising actinomycetes were evaluated for their physiological and plant growth-promotion properties under in vitro and in vivo conditions. All the isolates exhibited good growth at temperatures from 20 °C to 40 °C, pH range of 7–11 and NaCl concentrations up to 8%. Genome sequence of Streptomyces mangrovisoli MUSC 149T isolated from intertidal sediments BJM

ABSTRACT As the largest genus in Actinobacteria family, Streptomyces species have the ability to synthesize numerous compounds of diverse structures with bioactivities. Streptomyces mangrovisoli MUSC 149T was previously isolated as a novel streptomycete from mangrove forest in east coast of Peninsular Malaysia. The high quality draft genome of MUSC 149T comprises 9,165,825 bp with G + C content of 72.5%. Through bioinformatics analysis, 21 gene clusters identified in the genome were associated with the production of bioactive secondary metabolites. The presence of these biosynthetic gene clusters in MUSC 149T suggests the potential exploitation of the strain for production of medically important compounds.

Glucose(xylose) isomerase production by Streptomyces sp. CH7 grown on agricultural residues BJM

Streptomyces sp. CH7 was found to efficiently produce glucose(xylose) isomerase when grown on either xylan or agricultural residues. This strain produced a glucose(xylose) isomerase activity of roughly 1.8 U/mg of protein when it was grown in medium containing 1% xylose as a carbon source. Maximal enzymatic activities of about 5 and 3 U/mg were obtained when 1% xylan and 2.5% corn husks were used, respectively. The enzyme was purified from a mycelial extract to 16-fold purity with only two consecutive column chromatography steps using Macro-prep DEAE and Sephacryl-300, respectively.
