Over the last 10 years, DNA sequences of more than 600 bacterial species have been deposited in databases and are now available to search any gene, motif, or regulatory sequence of interest. Although genome data are instrumental in phylogenetic analysis and in silico design of metabolic and regulatory networks, only a very small fraction of the information has been experimentally validated. A striking example is lipoproteins predicted from genome sequences. Despite the predominance of this class of surface proteins in bacteria (up to 0.5 to 8% of the proteome), very few of these proteins have been identified as lipoproteins by biochemical methods (19). Analysis of myosin heavy chains at the protein level in horse skeletal muscle Inra

Combined methodologies of enzyme-linked immunosorbent assay (ELISA), sodium dodecyl sulphate polyacrilamide gel electrophoresis (SDS-PAGE), immunoblotting, traditional myofibrillar ATPase (mATPase) histochemistry and immunocytochemistry of whole biopsied samples were used to study myosin heavy chain (MHC) isoforms in the equine gluteus medius muscle. The ELISA technique allowed the quantification of the three MHC isoforms known to be present in different horse muscles: slow (MHC-I) and two fast (termed MHC-IIA and MCH-IIX). Biochemical and antigenic characterisation of Mycoplasma gallisepticum membrane proteins P52 and P67 (pMGA) Inra

Two membrane proteins from the avian pathogen Mycoplasma gallisepticum have been previously purified using a simple, efficient and non-denaturing method: a lipoprotein P67 (pMGA) and P52. In the current study, the lipid part of P67 was chemically analysed. The molecular structure of the lipoprotein-lipid component was determined to be S-glyceryl cysteine with two O-ester-linked acyl chains. Fatty acid analysis of the purified P67 indicated a heterogeneous composition: palmitic acid (16:0)>stearic acid (18:0)>oleic acid (18:1c)>myristic acid (14:0), with 16:0 as the major component. Caractérisation de Lactuca sativa L. et des espèces apparentées par électrofocalisation des estérases Inra

Les isoenzymes des estérases foliaires de Lactuca sativa et des espèces apparentées (L. aculeata, L. serriola, L. saligna, L. virosa) ont été séparées par électrofocalisation sur gel de polyacrylamide en gradient de pH 4 à 6,5. La variabilité des estérases est faible au sein de chaque espèce. L. sativa et L. serriola présentent des diagrammes très voisins qui confirment leur proche parenté ; L. aculeata apparaît proche de L. serriola ; L. saligna et L. virosa sont très distinctes des trois autres espèces. Les diagrammes d’estérases permettent de tester le caractère hybride des plantes issues de croisements interspécifiques (L. sativa x L. virosa).

Afin de mieux définir la fertilisation minérale des gazons, les exigences en éléments majeurs de 6 espèces de graminées à gazon : Lolium perenne L. cv. « Manhattan », Festuca rubra L. cv. « Manoir », Poa pratensis L. cv. « Monopoly », Phleum bertelonii L. cv. « Sport », Festuca arundinacea Schreb. cv. « Ludion ». Agrotis tenuis Sibth., ont été déterminées au moyen de courbes de réponse à l’azote, au phosphore et au potassium. Ces courbes de réponse sont obtenues à l’aide d’une technique de culture en solutions nutritives sur substrat inerte avec aération périodique automatique.  Replication defective retroviral vectors are regularly used for transfer and expression of exogenous genes into dividing cells and in animals. Since lentiviruses are able to infect terminally differentiated and non-dividing cells, their use to produce replication defective vectors may overcome this limitation. We developed two replication-defective lentiviral vectors based on the genome of Caprine Arthritis Encephalitis Virus (CAEV). The first vector (pBNL2) carries the neo and lacZ marker genes. Genome-wide analysis of the response to cell wall mutations in the yeast Saccharomyces cerevisiae Inra

Perturbations of the yeast cell wall trigger a repair mechanism that reconfigures its molecular structure to preserve cell integrity. To investigate this mechanism, we compared the global gene expression in five mutant strains, each bearing a mutation (i.e. fks1, kre6, mnn9, gas1, and knr4 mutants) that affects in a different manner the cell wall construction. Altogether, 300 responsive genes were kept based on high stringency criteria during data processing. Guard cell inward K+ channel activity in Arabidopsis involves expression of the twin channel subunits KAT1 and KAT2 Inra

Stomatal opening, which controls gas exchanges between plants and the atmosphere, results from an increase in turgor of the two guard cells that surround the pore of the stoma. KAT1 was the only inward K+ channel shown to be expressed in Arabidopsis guard cells, where it was proposed to mediate a K+ influx that enables stomatal opening. We report that another Arabidopsis K+channel, KAT2, is expressed in guard cells. More than KAT1, KAT2 displays functional features resembling those of native inward K+ channels in guard cells. Coexpression inXenopus oocytes and two-hybrid experiments indicated that KAT1 and KAT2 can form heteromultimeric channels. The data indicate that KAT2 plays a crucial role in the stomatal opening machinery.

Identification of new noncoding RNAs in Listeria monocytogenes and prediction of mRNA targets Inra

To identify noncoding RNAs (ncRNAs) in the pathogenic bacterium Listeria monocytogenes, we analyzed the intergenic regions (IGRs) of strain EGD-e by in silico-based approaches. Among the twelve ncRNAs found, nine are novel and specific to the Listeria genus, and two of these ncRNAs are expressed in a growth-dependent manner. Three of the ncRNAs are transcribed in opposite direction to overlapping open reading frames (ORFs), suggesting that they act as antisense on the corresponding mRNAs. The other ncRNA genes appear as single transcription units. One of them displays five repeats of 29 nucleotides. Identification of the sex Pheromone of Sesamia cretica Lederer Inra

By using solid phase micro-extraction and gas chromatography-mass spectrometry analyses, a sex pheromone blend for the stem borer, Sesamia cretica Lederer (Lepidoptera, Noctuidae), was identified as consisting of (Z)-9-tetradecen-1-ol (80%), (Z)-9-tetradecen-1-yl acetate (10%), and (Z)-11-hexadecen-1-ol (10%). The first two components had previously been discovered as attractants for S. cretica in field tests, but had not been identified in the female's sex pheromone gland. A field-trapping trial showed that the three-component blend gave the highest catches of male S. cretica. Rotaviruses are large, complex icosahedral particles consisting of three concentric capsid layers. When the innermost capsid protein VP2 is expressed in the baculovirus-insect cell system it assembles as core-like particles. The amino terminus region of VP2 is dispensable for assembly of virus-like particles (VLP). Coexpression of VP2 and VP6 produces double layered VLP. We hypothesized that the amino end of VP2 could be extended without altering the auto assembly properties of VP2.
