Anticorpos em bovinos (Bos indicus e Bos taurus) e bubalinos (Bubalus bubalis) inoculados com oocistos de Toxoplasma gondii. Estudo comparativo Arq. Bras. Med. Vet. Zootec.

Três animais de cada espécie (Bos indicus, Bos taurus e Bubalus bubalis) foram inoculados, via oral, com 2×10(5) oocistos de Toxoplasma gondii. Seis outros animais, dois de cada espécie, foram mantidos como testemunhas. A resposta de anticorpos avaliada por meio da reação de imunofluorescência indireta iniciou-se a partir do quinto dia pós-inoculação (DPI) nos zebuínos e bubalinos, e no sétimo DPI nos taurinos. Os títulos sorológicos nos taurinos permaneceram elevados até o final do experimento (70º DPI), alcançando níveis máximos (1:16.384) entre o 42º e 49º DPI. Nos zebuínos e bubalinos o maior título de anticorpos anti-Toxoplasma foi de 1:256. A resposta de anticorpos mais ou menos acentuada não está necessariamente relacionada à sensibilidade ao T....

The cross-reactive antigens of Neospora caninum and Toxoplasma gondii are important in the exploration to determine the common mechanisms of parasite-host interaction. In this study, a gene encoding N. caninum apical membrane antigen 1 (NcAMA1) was identified by immunoscreening of a N. caninum tachyzoite cDNA expression library with antisera. from mice immunized with recombinant T gondii apical membrane antigen 1 (TgAMA 1). NcAMA1 was encoded by an open reading frame of 1695 bp, which encoded a protein of 564 amino acids. The single-copy NcAMA1 gene was interrupted by seven introns. Characterization of a leucine aminopeptidase from Toxoplasma gondii OAK

The M17 family leucine aminopeptidase (LAP) hydrolyze amino acids from the N-terminus of peptides. Many LAPs from parasitic protozoa including Plasmmodium, Trypanosoma and Leishmania, have been intensely investigated because of their crucial roles in parasite biology. In this study, a functional recombinant Toxoplasma gondii LAP (rTgLAP) was expressed in Escherichia coli and its enzymatic activity against synthetic substrates for aminopeptidase, as well as the cellular localization was determined. Our results indicated that TgLAP is a functional aminopeptidase in the cytoplasma of T. gondii.

Characterization of a novel thrombospondin-related protein in Toxoplasma gondii OAK

Toxoplasma gondii is an obligate intracellular protozoan parasite that invades a wide range of host cells. Upon encountering host cells, the parasite releases a large variety of proteins from secretory organelles, such as micronemes, rhoptries and dense granules. The secretion of microneme protein is essential for parasite invasion. We found that a secreted protein with an altered thrombospondin repeat of Toxoplasma gondii (TgSPATR) was a novel microneme protein, which was different from known microneme proteins that carry thrombospondin repeat domains. TgSPATR was secreted in response to an intra-parasitic elevation of Ca2+ and probably secreted during early stages of parasite invasion. Detection of a high prevalence of antibodies against Toxoplasma gondii in cattle in Northern and Midwestern Brazil Rev Salud Anim.

The aim of this study was to determine the prevalence of Toxoplasma gondii in cattle from the Northern and Midwestern regions of Brazil. Serum samples were collected from 1789 animals and tested by indirect enzyme-linked immunosorbent assay (ELISA). The overall prevalence of T. gondii was 83.40% (1492/1789). The prevalence rates of T. gondii-seropositive animals observed in the states of Pará, Tocantins and, Mato Grosso were 87.45%, 87.79% and 73.06%, respectively. The detection of high prevalence rates of T. gondii in cattle deserves special attention because they are the main source of high biological value protein for humans. This finding indicates the need for further studies on the risk that these animals may pose to public health.

The Amazonian manatee (Trichechus inunguis) is an aquatic mammal that inhabits freshwater environments and is endemic to the Amazon Basin. The presence of Toxoplasma gondii antibodies was investigated in 19 manatees in one rescue unit in the northern region of Peru. Antibodies to T. gondii were detected in 12 (63.2 %) of 19 animals by using the modified agglutination test (titer, 1:25), and no association between sex and age of the animals and the presence of T. gondii antibodies was observed (p < 0.05). The results suggest a contamination by T. gondii oocysts in the aquatic environment where these animals live.

Background: Toxoplasma gondii is a highly prevalent protozoan that can infect all warm-blooded animals, including humans. Its definitive hosts are Felidae and its intermediate hosts include various other mammals and birds, including pigs. It is found in the meat of livestock which is a major source of human infection. Hence the control of toxoplasmosis in pigs is important for public health. Enzyme-linked immunosorbent assays using recombinant TgSAG2 and NcSAG1 to detect Toxoplasma gondii and Neospora caninum-specific antibodies in domestic animals in Turkey OAK

Considering the scarce information on occurrences of Toxoplasma gondii and Neospora caninum in domestic animals from Turkey, the aim of this study was to investigate the seroprevalence of these parasite infections in cattle, horses, sheep, goats and dogs in Turkey. The specific antibodies against T gondii and N. caninum were detected by iELISAs based on the recombinant TgSAG2 or NcSAG1 in a total of 2,039 serum samples from eleven provinces. The seroprevalence of T. gondii infections was 46.3%, 4.0%, 20.0%, 12.9% and 19.8%, that of N. caninum infections was 0.3%, 7.4%, 2.1%, 3.2% and 16.6% in the horses, cattle, sheep, goats and dogs, respectively. These results indicated that T gondii and N. caninum infections are prevalent in Turkish domestic animals.

Evaluation of novel oocyst wall protein candidates of Toxoplasma gondii OAK

Oocyst stage of Toxoplasma gondii is characterized by a durable wall that confers a strong protection to this protozoan parasite in face of harsh environmental conditions. Thus, it is considered the key for transmission of T. gondii. Analysis of oocyst wall composition is mandatory therefore; the aim of this study was to identify novel T. gondii oocyst wall proteins and test their use in detection of these oocysts in environmental samples. Five candidates of novel T. gondii oocyst wall proteins (TgOWPs) were identified and named TgOWP8 through TgOWP12. A gene encoding Babesia gibsoni thrombospondin-related adhesive protein (BgTRAP), known as a vaccine candidate, was stably expressed in Toxoplasma gondii (Tg/BgTRAP). The molecular weight and the antigenic reaction of recombinant BgTRAP expressed by the Tg/BgTRAP were similar to the original ones expressed by B. gibsoni. To evaluate the antigenicity of the recombinant BgTRAP expressed by T. gondii, the lysates of the recombinant parasite tachyzoites were intraperitoneally injected into mice. The serum collected from Tg/BgTRAP-immunized mouse could react to B. gibsoni parasites, while the serum collected from wild-type T. gondii tachyzoites (Tg/wt)-immunized mice did not.
