In sexually reproducing organisms, primordial germ cells (PGCs) give rise to the cells of the germ line, the gametes. In many animals, PGCs are set apart from somatic cells early during embryogenesis. This study explores the origin of primordial germ cells (PGCs) of the zebra fish and examines their morphology during early development (1st day-15th day). PGCs were selectively stained by the alkaline phosphatase histochemical reaction and viewed by light and electron microscopy from the time they are first detectable in the yolk sac endoderm. PGCs occurred in the subendodermal space on the syncytial periblast; differing from the surrounding endodermal cells. An ultrastructural study of spermiogenesis in two species of Sitophilus: Coleoptera: Curculionidae Biocell

The spermiogenesis of Sitophilus zeamais and Sitophilus oryzae, the maize and the rice weevil, respectively, was studied by light microscopy and scanning and transmission electron microscopy. Sitophilus spp. is the most widespread and destructive primary pest of stored cereals in the world. The spermiogenesis occurs within cysts. There are approximately 256 germ line cells per cyst. Inside each cysts, all the spermatids are in the same stage of maturation. The ultrastructure of the spermatozoa of S. zeamais and S. oryzae is similar to that described for other beetles. Análise ultra-estrutural de folhas de três espécies de Solanaceae após inoculação com o vírus da necrose branca do tomateiro (VNBT - Tymovirus) Rev. Bras. Bot.

Foi estudada a ultra-estrutura foliar das solanáceas Nicotiana glutinosa L., Lycopersicon pimpinellifolium L. e Physalis angulata L. inoculadas com o vírus da necrose branca do tomateiro (VNBT - Tymovirus). As plantas mantidas em casa-de-vegetação com temperatura constante de 25 °C foram inoculadas quando apresentavam três a quatro folhas totalmente expandidas. Quinze dias após a inoculação, foram coletadas amostras do terço médio do limbo da 3ª ou da 4ª folha a partir do ápice. As amostras foram preparadas para análise em microscopia eletrônica de transmissão segundo técnicas convencionais. Approche ecophysiologigue et ultrastructurale de l'osmoregulation chez les crustaces decapodes ArchiMer

This paper deals mostly with recent physiological data on ion transport across the gill tissue and an attempt is made to correlate them with blood osmoregulation and ecological distribution in euryhaline decapods. A particular attention is paid to the gills of the fully euryhaline Chinese crab Eriocheir sinensis . Supporting this ecophysiological approach, there is an important ultrastructural analysis of the gill epithelium under various salinity conditions. The possible part played by the cuticle in ions movements is also considered.

Aspects ultrastructuraux de la biodegradation de l''ancienne cuticule de Carcinus maenas durant la periode preecdysiale ArchiMer

Ultrastructural observations of various integumental regions of the crab Carcinus maenas show an important biodegradation of the old sclerites during premolt. At the ecdysis, this biodegradation extends from the basis of the membranous layer up the upper third of the principal layer. The first biodegradation features are a disorganization followed by a complete disappearance of the microfibrils. Numerous ecdysial droplets and double membrane bounded microstructures probably take part in biodegradation processes.

Bonamia sp. is a pathogenic parasite that occurs in the haemocytes of dredge oysters Ostrea chilensis Philippi in New Zealand, Ultrastructurally it resembles other haplosporidians in the possession of haplosporosomes, haplosporogenesis, persistence of mitotic microtubules during interphase and of the nuclear envelope during mitosis, and occurrence of a diplokaryotic or multi-nucleate plasmodial stage. Another stage containing a large vacuole derived from enlargement of 1 or more mitochondria has not previously been described from other haplosporidians. Caracterisation des hemocytes d''un mollusque bivalve marin, la nacre, Pinna nobilis L. 1758 ArchiMer

The study of tinctorial affinities, in vitro behaviour and ultrastructure of the hemocytes of Pinna nobilis allows us to group them into two main families: basophilic, neutrophilic and acidophilic granulocytes; hyalinocytes, three categories can be distinguished among these cells: small hyalinocytes, hyalinocytes containing smooth endoplasmic reticulum and the macrophage population. The hemocytes participate in the regulation of metabolism by the storage and degradation of glycogen. They contribute to the detoxification process by the sequestration of mineral elements in their lysosomes. Their role in antibacterial defense is suggested by their bacterian phagocytic activity.

Caracteristiques ultrastructurales du spermatozoide de la coquille Saint-Jacques, Pecten maximus (Mollusque, Bivalve) ArchiMer

The spermatozoon of Pecten maximus is of the primitive type usually associated with species having external fertilization or emitting the sperm in the water. The size of the nucleus is reduced. The middle piece is similar to that reported for several Bivalves. It contains glycogen. But the spermatozoon of P. maximus exhibits its own ultrastructural characteristics: the short acrosome differs in size, shape, position and ultrastructure or by a combination of these from that of many bivalves. No axial rods was observed in contrast to several species of Mytilidae, Veneridae and Ostreidae. These originalities could go to the genus level.

Changes in sperm features during the movement phase are especially interesting to study in external fertilization species whose sperm duration movement is long because this implies a significant adaptation of moving cells to the external medium. This study describes the changes in tetraploid Pacific oyster sperm characteristics in relation to time post activation. Sperm individually collected on three tetraploid males were activated in seawater. The ultrastructure of male copulatory organs having a stylet has been studied in some genera of the Proseriata. Within the Monocelididae there was a variety of stylet-like hard structures. The stylet in <i>Monocelis fusca</i> was a differentiation of the basement membrane of the epithelium lining a penis-like muscular papilla. The penis papilla in <i>Ectocotyla</i>consisted of circular muscles surrounded by a thickened basement membrane and an epithelium. <i>Archilopsis sp.</i> and <i>Archilina sp.</i> with a duplex copulatory bulb, had a stylet within a spiny cirrus. The aim of our study was to compare the nephrotoxic effects of liposomal Amphotericin B (Ambisome) and Amphotericin B lipid complex (Abelcet) on rat kidneys at short (14 days) and long term (28 days) treatment applications. Thirty-six male Wistar rats were included and divided into six groups (n=6). Groups 1 and 4 are composed as control groups by administrating intraperitoneal (ip) 0, 9 molar Serum physiologic for a period of 14 and 28 days respectively. Group 2 and 3 are treated with 5 mg/kg Ambisome and 5 mg/kg Abelcet for 14 days respectively, Group 5 and 6 are treated with same agents for 28 days respectively. To assess relationships between neuropeptide-binding sites and receptor proteins in rat brain, the distribution of radioautographically labeled somatostatin and neurotensin-binding sites was compared to that of immunolabeled sst2A and NTRH receptor subtypes, respectively. By light microscopy, immunoreactive sst2A receptors were either confined to neuronal perikarya and dendrites or diffusely distributed in tissue. Background The effects of exposure to copper, during growth, on the production of biomass, total protein, catalase, glutathione-S transferase, glutathione peroxidase, peroxidase, polyphosphate, acid and alkaline phosphatases, ultrastructure and the ability to remove this metal from Aspergillus niger, obtained from caatinga soil, were evaluated. Results All parameters tested were influenced by the concentration of metal in the culture medium. Cytochemical localization of acid phosphatase in Stigeoclonium tenue (Chaetophorales, Chlorophyceae) Biocell

Nonspecific acid phosphatases are a group of enzymes whose activity increases the availability of exogenous and endogenous orthophosphate either through extra- or intracellular hydrolysis of phosphate compounds. Our study demonstrates the activity of acid phosphatases in the filamentous freshwater alga Stigeoclonium tenue. These enzymes were detected following a cerium-based method in which cerium was used as an orthophosphate-capture reagent. In thalli from S. tenue from the natural environment, acid phosphatases were found in the longitudinal cell wall, plasmalemma, and vacuole. The present ultrastructural study on developing and regenerating feathers of chick and zebrafinch describes the ultrastructural changes that occur during the differentiation of barb cells that leads to the formation of the ramus of barbs. Differently from barbule and barb cortical cells that accumulate feather keratin, barb medullary cells undergo to lipid degeneration. Eventually, lipids disappear and medullary cells become empty cavities in the central part of the ramus. The macroscopic ovaric stages of pink cuskeel (Genypterus blacodes) females from the Argentine Sea (SW Atlantic) are described. They were classified in five stages of development based on changes in the appearance and size of the ovary: juvenile (I), developing (II), spawning (III), spent (IV) and resting (V).

Development, comparative morphology and cornification of reptilian claws in relation to claws evolution in tetrapods Naturalis

The development of claws in different reptiles and their cornification are analyzed using histological, ultrastructural and autoradiographic methods. Claws develop at the tip of digits in relation to the growth of the terminal phalanx and appear as modified scales. The apical epidermis of digit becomes thickened and is associated with a mesenchymal condensation or a dense mesenchyme. The dorsal side of the digit becomes the unguis while the ventral side becomes the sub-unguis. The corneous layer in the unguis is thicker than in the sub-unguis and accumulates hard-keratin while corneocytes remain separated or partially fused. Bundles of hard-keratin tend to accumulate in parallel orientation with respect to the surface and are directed toward the claw tip....

Effect of Cigarette Smoking on the Structure of Hepatocytes: TEM Study International Journal of Morphology

The liver is one of the major organs that is indirectly affected by cigarette smoke. The aim of this project is to define the histologic and ultrastructural changes in normal liver cells after exposing animals to cigarette smoke. Thirty albino rats were exposed to cigarette smoke for 90 days, followed by morphologic examination of their livers under light microscope and electron microscope. The liver cells of cigarette smoke exposed rats showed mild swelling with increased eosinophilia. Ultrastructural examination of these cells demonstrated cytoplasm with highly proliferated and crowded mitochondria. There were many electron dense mitochondria. In the three species Dicentrarchus labrax, Sparus auratus , and Anguilla anguilla an artificial feeding (dry pellets) disturbs liver cells as revealed at ultrastructural level. In hepatocytes, anomalies consist of lipids storage (liposomes) considered as deviation in fatty acids metabolism. Moreover are observed cellular alterations (mitochondrial vacuolization, nuclear pycnosis) related to degenerative process. It is of great interest to understand wether liver modifications are just a reaction to an unbalanced feeding or a true trophic pathology.
