Efficiency of methods to detect Sclerotinia sclerotiorum in commercial soybean seed lots Rev. bras. sementes
Henneberg,Luciane; Jaccoud Filho,David de S.; Ruaro,Lucimeris; Panobianco,Maristela.
Most soybean pathogens are seed transmitted, deserving emphasis the fungus Sclerotinia sclerotiorum, which has been presenting worrying levels of field incidence in some soybean cropping areas in several Brazilian states. The objective of this study was to verify the efficiency of different methods for detecting S. sclerotiorum on soybean seeds artificially infected in the laboratory and from field production areas with a historical disease incidence. Seed samples of seven different cultivars collected from naturally infested fields, and one seed sample artificially inoculated in the laboratory were used. The following detection methods recommended in the literature were compared: Blotter test at 7 ºC, 14 ºC, and 21 ºC; Rolled Paper; and Neon-S. Results...

Endophytic fungi from Brachiaria grasses in Brazil and preliminary screening of Sclerotinia sclerotiorum antagonists Scientia Agricola
Gama,Danielly de Souza; Santos,Ítalo Augusto Férrer Melo; Abreu,Lucas Magalhães de; Medeiros,Flávio Henrique Vasconcelos de; Duarte,Whasley Ferreira; Cardoso,Patrícia Gomes.
ABSTRACT: Fungal endophytes of Brachiaria, a nonhost of Sclerotinia sclerotiorum, may harbor species with antagonistic effects against this plant pathogen. The objective of this work was to investigate the diversity of endophytic fungi associated with different Brachiaria species and hybrids and evaluate their potential to inhibit the plant pathogen S. sclerotiorum. Stem samples from 39 Brachiaria spp. plants were collected in pasture fields and experimental areas of three states of Brazil resulting in 74 endophytes isolated. Twenty-eight species were identified by sequences of the Internal Transcribed Spacer (ITS) and 18S rDNA regions. Paraconiothyrium sp. was the most abundant endophyte, accounting for 24 % (14 isolates) of total, and it was isolated...

Koga,Lucimara Junko; Bowen,Charles Roger; Godoy,Claudia Vieira; Oliveira,Maria Cristina Neves de; Hartman,Glen Lee.
The objective of this work was to evaluate the genetic diversity among Sclerotinia sclerotiorum isolates from Brazil and the USA, assess their aggressiveness variability, and verify the existence of an isolate-cultivar interaction. Isolate variability was determined by mycelial compatibility grouping (MCG), and isolate aggressiveness by cut-stem inoculations of soybean cultivars. Two experiments for MCGs and two for aggressiveness were conducted with two sets of isolates. The first set included nine isolates from the same soybean field in Brazil and nine from the Midwest region of the USA. The second set included 16 isolates from several regions of Brazil and one from the USA. In the first set, 18 isolates formed 12 different MCGs. In the second set, 81%...

New apiaceous hosts of Sclerotinia sclerotiorum in the Cerrado region of Brazil Horticultura Brasileira
Reis,A; Nascimento,WM.
This paper reports for the first time in the Cerrado region of Brazil the occurrence of white mold caused by Sclerotinia sclerotiorum in coriander (Coriandum sativum), parsley (Petroselinum crispum) and Peruvian carrot (Arracacia xanthorrhiza). The disease was observed on coriander, in Cristalina, Goiás State, on parsley in Gama, the Federal District and on Peruvian carrot in Ibiá, Minas Gerais State. Pathogenicity tests demonstrated that the fungus S. sclerotiorum is the causal agent of the observed symptoms in these plants. The three isolates obtained from naturally infected plants were inoculated in coriander (cv. Verdão), parsley (cv. Lisa Gigante), Peruvian carrot (cv. Amarela de Senador Amaral) and carrot (cv. Forto Nantes) and they were pathogenic...

New isolates of Trichoderma antagonistic to Sclerotinia sclerotiorum Biota Neotropica
Marques,Eder; Martins,Irene; Cunha,Mariana de Oliveira Cardoso; Lima,Marcello Arrais; Silva,João Batista Tavares da; Silva,Joseane Padilha da; Inglis,Peter Ward; Mello,Sueli Correa Marques.
Abstract Forty-nine isolates of Trichoderma from the Brazilian Midwest were evaluated for their antagonistic activity in vitro against Sclerotinia sclerotiorum (causal agent of white mold), which were then identified based on their nuclear ribosomal ITS sequences. Paired culture tests showed that all isolates exhibited some antagonism, with a maximum of 77% mycelial inhibition and complete inhibition of sclerotia production. Two isolates were found to be the most promising biocontrol agents, considering both antagonistic parameters (CEN1253 - T. koningiopsis and CEN1265 - T. brevicompactum). Five different species were identified: T. harzianum (23), T. spirale (9), T. koningiopsis (8), T. brevicompactum (7) and T. asperellum (2). These isolates are stored...

Potential of antimicrobial volatile organic compounds to control Sclerotinia sclerotiorum in bean seeds PAB
Fialho,Maurício Batista; Moraes,Maria Heloisa Duarte de; Tremocoldi,Annelise Roberta; Pascholati,Sérgio Florentino.
The objective of this work was to evaluate the potential of an artificial mixture of volatile organic compounds (VOCs), produced by Saccharomyces cerevisiae, to control Sclerotinia sclerotiorum in vitro and in bean seeds. The phytopathogenic fungus was exposed, in polystyrene plates, to an artificial atmosphere containing a mixture of six VOCs formed by alcohols (ethanol, 3-methyl-1-butanol, 2-methyl-1-butanol and phenylethyl alcohol) and esters (ethyl acetate and ethyl octanoate), in the proportions found in the atmosphere naturally produced by yeast. Bean seeds artificially contamined with the pathogen were fumigated with the mixture of VOCs in sealed glass flasks for four and seven days. In the in vitro assays, the compounds 2-methyl-1-butanol and...

Protocol adjustment improves the extraction of high-quality total RNA from common bean stems infected by Sclerotinia sclerotiorum Ciência e Agrotecnologia
Miranda,Rafael Novais de; Silva,Caroline Marcela da; Porto,Antonio Carlos da Mota; Pereira,Welison Andrade.
ABSTRACT The Straw Test is an assay developed to evaluate the resistance of common bean to white mold, in which the plant stems are inoculated and the symptoms of the disease are monitored. It is plausible to admit that investigating gene expression in pathogen-infected tissues may be strategically interesting. However, obtaining a quality RNA is a basic requirement for this purpose. Therefore, the objective of this study was to evaluate adjustments in protocols of commercial kits in the expectation of improving the quality of RNA obtained from bean stems. For this, plants of two lines were inoculated and the stems pathogen-infected were collected 72 hours after. For RNA extraction, two commercial reagents were used following the manufacturer’s...

Rapid PCR-based assay for Sclerotinia sclerotiorum detection on soybean seeds Scientia Agricola
Grabicoski,Edilaine Mauricia Gelinski; Jaccoud Filho,David de Souza; Pileggi,Marcos; Henneberg,Luciane; Pierre,Marcelo Luiz Cunha; Vrisman,Cláudio Mauricio; Dabul,Audrei Nisio Gebieluca.
Caused by Sclerotinia sclerotiorum, white mold is an important seed-transmitted disease of soybean (Glycine max). Incubation-based methods available for the detection and quantification of seed-borne inoculum such as the blotter test, paper roll and Neon-S assay are time-consuming, laborious, and not always sensitive. In this study, we developed and evaluated a molecular assay for the detection of S. sclerotiorum in soybean seeds using a species-specific PCR (polymerase chain reaction) primer set and seed soaking (without DNA extraction) for up to 72 h. The PCR products were amplified in all the samples infected with the pathogen, but not in the other samples of plant material or the other seed-borne fungi DNA. The minimum amount of DNA detected was 10 pg,...

Resistance of cotton genotypes to Sclerotinia sclerotiorum by the straw and oxalic acid tests PAB
Aguiar,Leandro Henrique Mundim; Sousa,Larissa Barbosa de; Coelho,Lísias; Lemes,Ernane Miranda; Castro,Leonardo Humberto Silva e; Cardoso,Daniel Bonifácio Oliveira.
Abstract: The objective of this work was to evaluate the efficiency of the straw and oxalic acid tests to identify resistance levels of white and colored cotton (Gossypium hirsutum) genotypes to white mold (Sclerotinia sclerotiorum). Ten genotypes were evaluated: five with colored fiber, four with white fiber, and a white-fiber susceptible genotype. The genotypes MAB-1 with white fiber and MAC-2 with colored fiber were the most resistant to white mold, according to the immersion in oxalic acid and straw tests, respectively. These genotypes can be recommended as resistance sources for breeding programs. Both assessed tests are complementary to each other; however, the straw test is more efficient in evaluating the resistance of cotton genotypes to white mold.

Selection of Trichoderma spp. strains for the control of Sclerotinia sclerotiorum in soybean PAB
Haddad,Patrícia Elias; Leite,Luis Garrigós; Lucon,Cleusa Maria Mantovanello; Harakava,Ricardo.
Abstract: The objective of this work was to evaluate, in vitro and in vivo, the potential of Trichoderma spp. strains to control Sclerotinia sclerotiorum in soybeans (Glycine max) and to perform the molecular identification of the best perfoming strains. The effect of 120 strains of Trichoderma spp. on the viability of S. sclerotiorum sclerotia was evaluated in vitro through immersion in suspension of conidia from the antagonists and plating in culture medium. The best performing strains were evaluated in vivo, in a greenhouse, for control of the pathogen inoculated on 'Pintado' soybean seeds and plants. Of the 120 strains tested in vitro, 22 strains of Trichoderma spp. caused 100% inhibition of sclerotia germination. In the greenhouse, five strains...
