Anhydrobiosis in yeast: activation effect BJMBR
Intracellular substances released into the medium during rehydration of dry yeast cells can significantly improve the quality of a synthetic medium. Acceleration of yeast growth in this medium and increased yield of biomass are observed simultaneously. The change in the molecular arrangement of intracellular membranes as a result of the strong dehydration of live organisms is a negative phenomenon that reduces the level of cell viability. However, this phenomenon also represents an adaptive mechanism which facilitates the maintenance of population viability as a whole under extreme environmental conditions

Autochthonous yeast populations from different brazilian geographic indications. Repositório Alice
SILVA, G. A. da; AGUSTINI, B. C.; MELLO, L. M. R. de; TONIETTO, J..
Yeasts are versatile microorganisms which show heterogeneity in their abilities of aromatic molecules formation. The metabolic conversions may improve the production of a particular compound already formed by the microorganism or promote the production of a completely new biochemicals. These conversions depend on the environment. The microbiome of terroir is unique. If the term terroir is a set of physical properties of a vineyard that contribute to the specific characteristics of its wine, the microorganisms will undoubtedly form an integral part of this concept. There are yeasts, filamentous fungi and bacteria that can affect the quality of the wine. The aim of the present study was to identify the autochthonous yeast populations of grape berries...

Baker's yeast as a potential substitute for live algae in aquaculture diets: Artemia as a case study OMA
Coutteau, P.; Lavens, P.; Sorgeloos, P..
In this study baker's yeast was evaluated as a substitute for live Dunaliella tertiolecta algae in the culture of the brine shrimp Artemia . Consumption of fresh baker's yeast resulted in poor growth and survival of brine shrimp. However, the nutritional value of the yeast significantly improved after complete removal of the yeast cell wall by enzymatic treatment. Baker's yeast was also made digestible for Artemia by simple chemical treatment which did not reduce rigidity of the yeast cell. The external mannoprotein layer of the yeast cell wall is probably the major barrier to digestion by Artemia . Chemically treated baker's yeast offers promising possibilities as a substitute for...

Abstract: This work aimed to evaluate nine strains of yeast, previously identified as good producers of lipids in honey medium, for selecting the most suitable strain for the production of lipids in cheese whey medium and compared two well known extraction methods of lipids from the culture medium. The highest yield of total lipids was 1.27 g.L-1 produced by Cryptococcus laurentii 11. A comparison was made between the two culture media: cheese whey and liquid YEPG, and two lipid extraction methods: Bligh and Dyer and Folch et al. for C. laurentii. The experiments were performed with 22 full factorial design using two factors and two levels. Lipid content was higher in cheese whey and there was no difference in the extraction methods statistically. The...

Candida spp was isolated from 59 (68.60%) out of eighty six samples of oral mucosa of AIDS patients. The identification, based or the production of a germ tube and chlamydospores, and on the assimilation and fermentation of carbohydrates, revealed 52 strains (88.13%) of C. albicans, 4 (6.77%) of C. tropicalis and 3 (5.08%) of C. krusei. The susceptibility of these strains to amphotericin B, flucytosine, itraconazole, fluconazole and ketoconazole was determined using the agar dilution method. Comparing the minimum inhibitory concentration values found in the susceptibility test with the serum levels achieved by these drugs, only 8.47% and 5.08% of the yeasts strains proved to be resistant to amphotericin B and flucitosyne, respectively. A high frequency of...

Evaluation of MALDI-TOF mass spectrometry for identification of environmental yeasts and development of supplementary database. Repositório Alice
AGUSTINI, B. C.; SILVA, L. P. da; BLOCH JUNIOR, C.; BONFIM, T. M. B.; SILVA, G. A. da.
Yeast identification using traditional methods which employ morphological, physiological, and biochemical characteristics can be considered a hard task as it requires experienced microbiologists and a rigorous control in culture conditions that could implicate in different outcomes. Considering clinical or industrial applications, the fast and accurate identification of microorganisms is a crescent demand. Hence, molecular biology approaches has been extensively used and, more recently, protein profiling using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) has proved to be an even more efficient tool for taxonomic purposes. Nonetheless, concerning tomass spectrometry, data available for the differentiation of...

An electronic nose (E-nose) coupled to gas chromatography was tested to monitor alcoholic fermentation by Saccharomyces cerevisiae ICV-K1 and Saccharomyces cerevisiae T306, two strains well-known for their use in oenology. The biomass and ethanol concentrations and conductance changes were measured during cultivations and allowed to observe the standard growth phases for both yeast strains. The two strains were characterized by a very similar tendency in biomass or ethanol production during the fermentation. E-nose was able to establish a kinetic of the production of aroma compounds production and which was then easy to associate with the fermentation phases. Principal Component Analysis (PCA) showed that the data collected by E-nose during the...

EN-US: The Brazil nut is an important commodity that grows in the Amazon rainforest and is notable for its high nutritional value. However, the effect that packaging has on product shelf-life is unclear. This study evaluated the microbiological and nutritional quality of unshelled Brazil nuts packed in (a) nylon and polypropylene under vacuum, and (b) in polyethylene terephthalate and polyethylene aluminized films. The nuts were stored for nine months under natural environmental conditions. The most probable number of total and thermotolerant coliforms and Escherichia coli, and counting of molds and yeast were determined in the unshelled nuts shortly after collection and every three months during storage. Moisture, lipids, ash, neutral detergent fiber, and...

Isolation and identification of yeasts and filamentous fungi from yoghurts in Brazil BJM
Moreira,Silvia Regina; Schwan,Rosane Freitas; Carvalho,Eliana Pinheiro de; Wheals,Alan E..
Seventy-two cartons of yoghurt were sampled three times at monthly intervals from four different local manufacturers. Total counts were close to 6 x 10(7) cells g-1 of yoghurt. Yeast counts varied from 1 to 2,700 g-1. There was no evidence of systematic contamination at source but this longitudinal study revealed that ad hoc contamination and improper storage led to the higher yeast counts. Contamination was generally higher in the hotter months but was lower overall than reported from other countries. A total of 577 yeast isolates were identified belonging to ten species. The most abundant yeasts were, in order, Debaryomyces hansenii, Saccharomyces cerevisiae, Mrakia frigida, Hansenula spp., Candida parapsilosis, Debaryomyces castellii and Candida...

Marine culturable yeasts in deep-sea hydrothermal vents: species richness and association with fauna ArchiMer
Burgaud, Gaetan; Arzur, Danielle; Durand, Lucile; Cambon-bonavita, Marie-anne; Barbier, Georges.
Investigations of the diversity of culturable yeasts at deep-sea hydrothermal sites have suggested possible interactions with endemic fauna. Samples were collected during various oceanographic cruises at the Mid-Atlantic Ridge, South Pacific Basins and East Pacific Rise. Cultures of 32 isolates, mostly associated with animals, were collected. Phylogenetic analyses of 26S rRNA gene sequences revealed that the yeasts belonged to Ascomycota and Basidiomycota phyla, with the identification of several genera: Rhodotorula, Rhodosporidium, Candida, Debaryomyces and Cryptococcus. Those genera are usually isolated from deep-sea environments. To our knowledge, this is the first report of yeasts associated with deep-sea hydrothermal animals.
